Thymosin alpha-1 is a 28-amino acid acetylated peptide corresponding to the N-terminal fragment of prothymosin alpha, an endogenous thymic peptide. In vitro and rodent studies examined its association with activation of dendritic cells via Toll-like receptor signaling, including TLR2, TLR3/4/9, and TLR9-dependent pathways linked to downstream IRF3 and NF-κB signaling and induction of indoleamine 2,3-dioxygenase (IDO) expression. A study in human monocyte-derived macrophages examined its association with complement receptor-mediated phagocytosis. A rodent model of antifungal immune signaling examined its association with dendritic cell priming for Th1-associated cytokine responses via TLR9.
